Plug-and-play assembly of biodegradable ionizable lipids for potent mRNA delivery and gene editing in vivo

This blog discusses the development of a plug-and-play assembly strategy for the combinatorial synthesis of biodegradable ionizable lipids (ILs) recently published by researchers (Han et al.) at the University of Pennsylvania. Compared to benchmark ILs, the lead IL, synthesized in one step from an amine and a dialkyl maleate, demonstrates superior performance in delivering various CRISPR mRNA therapeutics in wild-type and transgenic mice.

By introducing various modifications through chemical or enzymatic methods across the entire mRNA sequence—from cap to tail—translational activity can be enhanced, and immunogenicity risks reduced. To that end, scientists at TriLink BioTechnologies have developed a novel modification to the poly(A) tail, which further improves the stability and translational activity of RNA therapeutics.
