Guiding Innovation: The Roles of Biomarkers in CAR-based Therapies


Chimeric Antigen Receptor (CAR) cell therapies are reshaping cancer treatment, particularly in hematologic malignancies where CAR-T therapies have demonstrated durable remissions in patients with otherwise limited options. The field continues to evolve with the rise of CAR-NK therapies, offering new possibilities for solid tumor targeting and allogeneic, off-the-shelf applications.

Alongside these innovations, biomarkers have emerged as essential tools to support manufacturing consistency, enhance safety monitoring, and improve therapeutic outcomes. Measuring What Matters: The Functional IL-15/IL-15Ra Complex


The IL-15/IL-15Rα complex plays a pivotal role in regulating immune responses by activating NK cells and memory CD8+ T cells. Unlike free IL-15, the complex reflects the cytokine’s biologically active form in vivo, making it a more accurate indicator of immune activity. In healthy individuals, this complex is tightly regulated and typically found at low levels, serving as a baseline for understanding immune homeostasis.

PBL’s VeriKine-HS Total Human IL-15 ELISA (41702-1) and three competitors’ standards were assayed on their respective ELISAs. PBL’s ELISA shows 1 to 2 logs improved sensitivity

Accurate measurement of the IL-15/IL-15Rα complex in serum is essential for detecting immune dysregulation in diseases such as autoimmunity, cancer, and chronic inflammation. It also holds value as a pharmacodynamic biomarker in IL-15-based therapies, helping researchers monitor therapeutic response and optimize dosing. Given its low abundance, sensitive and specific assays are required to capture meaningful data that can guide both clinical research and translational applications.


PBL’s VeriKine-HS Total Human IL-15 ELISA Kit (41702-1) is designed for high-sensitivity measurement of both free IL-15 and the biologically active IL-15/IL-15Rα complex, offering reliable quantification down to sub-pg/mL levels. This dual-detection capability makes it an ideal tool for researchers studying immune regulation, disease biomarkers, or therapeutic responses involving IL-15 signaling.
